Abstract

There is provided a pharmaceutical preservative free composition suitable for topical administration to an eye, the composition comprising (a) an oxazolidinone antimicrobial drug, for example linezolid, in a therapeutically or prophylactically effective drug concentration that is above the practical limit of solubility of the drug in a substantially isotonic aqueous solution at a physiologically compatible pH, and (b) a pharmaceutically acceptable cyclodextrin compound in a concentration sufficient to maintain the drug in solution at such a drug concentration. The composition is particularly useful for the treatment and/or prevention of eye infections due to gram positive bacteria.

Claim

What is claimed is: 1. A pharmaceutical composition suitable for topical administration to an eye, comprising: (a) an oxazolidinone antibiotic drug in a concentration effective for treatment and/or prophylaxis of a gram-positive bacterial infection of at least one tissue of the eye, the concentration being above the practical limit of solubility of the drug in an aqueous solution at a physiologically compatible pH; and (b) a pharmaceutically acceptable cyclodextrin compound in a cyclodextrin concentration sufficient to maintain the drug in solution at the drug concentration, wherein, the pharmaceutical composition is preservative free. 2. The composition of claim 1 wherein the oxazolidinone antibiotic drug is a compound of formula wherein: R1 is selected from (a) H, (b) C1-8 alkyl optionally substituted with at least one F, Cl, OH, C1-8 alkoxy, and C1-8 acyloxy or C1-8 benzoxy, including a C3-6 cycloalkyl group, (c) amino, (d) mono- and di(C1-8 alkyl)amino and (e) C1-8 alkoxy groups; R2 and R3 are independently selected from H, F and Cl groups; R4 is H or CH3; R5 is selected from H, CH3, CN, CO2R1 and (CH2)mR6 groups, where R1 is as defined above, R6is selected from H, OH, OR1, OCOR1, NHCOR1, amino, mono- and di(C1-8 alkyl)amino groups, and m is 1 or 2; n is 0, 1 or 2; and X is O, S, SO, SO2, SNR7 or S(O)NR7 where R7 is selected from H, C1-4 alkyl (optionally substituted with one or more F, Cl, OH, C1-8 alkoxy, amino, C1-8 mono- or di(C1-8 alkyl)amino groups), and p-toluenesulfonyl groups; or a pharmaceutically acceptable salt thereof. 3. The composition of claim 2 wherein, in said formula, R1 is CH3; R2 and R3 are independently selected from H and F but at least one of R2 and R3 is F; R4 and R5 are each H; n is 1; and X is selected from O, S and SO2. 4. The composition of claim 1 wherein the oxazolidinone antibiotic drug is selected from the group consisting of: linezolid, eperezolid, N-((5S)-3-(3-fluoro-4-(4-(2-fluoroethyl)-3-oxopiperazin-1-yl)phenyl)-2-oxooxazolidin-5-ylmethyl)acetamide, (S)-N-[[3-[5-(3-pyridyl)thiophen-2-yl]-2-oxo-5-oxazolidinyl]methyl]acetamide, (S)-N-[[3-[5-(4-pyridyl)pyrid-2-yl]-2-oxo-5-oxazolidinyl]methyl]acetamide hydrochloride and N-[[(5S)-3-[4-(1,1-dioxido-4-thiomorpholinyl)-3,5-difluorophenyl]-2-oxo-5-oxazolidinyl]methyl]acetamide. 5.

Also, a method for providing a high build, fast drying and fast hardening aqueous coating at ambient conditions is provided.

Abstract

In a machine for dispensing stackable roofing washers individually and for driving fasteners through such washers, a shuttle is actuatable to displace a lowermost washer of one from a stack, either a small, circular or large, square washer. A gate is biased toward a lowermost position wherein it provides sufficient clearance for the lowermost washer but not any overlying washer to pass beneath it. A pawl mounted pivotally to the gate is biased to a lowermost position wherein the pawl engages a small washer displaced by the shuttle so as to restrain it against backward movement. A stop is adjustable so as to limit movement of a washer of whichever size is being displaced. A measuring bar is mounted via the stop. A screw gun, which provides a primary handle, and a secondary handle are mounted to an upright element so as to be independently adjustable.

Abstract

A roofing washer-dispensing machine having improved features is disclosed. A novel magazine comprises a substantially box-like container with a substantially open face, for housing a stack of circular roofing washers having inserts, and a substantially rigid rod removably mounted within or upon to a top wall of the box so as to extend downwardly into the central apertures of the stack washers, except for the lowermost washer, so as to restrain the remaining washers. The machine comprises a shuttle arranged to displace the lowermost washer from the stack when it is desired to dispense such washer. The shuttle includes a novel arrangement of means overlying and underlying marginal portions of such washer.
